Summary
Polymerase chain reaction (PCR) is a well-established method for nucleic acid amplification. However, it has limitations in many situations, such as amplifications of large genomes or heterogenous templates containing multiple DNA/cDNA species. Another amplification method, multiple displacement amplification (MDA), based on ph29 DNA polymerase, has been developed as the gold standard for single-cell and globe gene expression analysis. MDA outperforms PCR in terms of amplification bias, sensitivity and fidelity. But it has an issue with generation of non-template product, or junk DNA. This invention relates to the development of a special primers and kits that significantly reduce generation of junk DNA and enhance the power and potential of MDA.
