Summary
This invention relates to hydroxylated tropolone inhibitors of nucleotidyl1-transferase superfamily enzymes that suppress replication of HSV- and HSV-2. The compounds disclosed appear to inhibit a different enzymatic activity than the existing nucleoside analog anti-herpes drugs. They do so with a striking capacity to suppress virus replication at very low toxicity to uninfected cells. Also disclosed are novel RNAseH inhibitors that work synergistically with nucleoside analogs against Hepatitis B viral replication because the two classes of drugs target physically distinct active sites on the viral polymerase protein.
